The Amino Acid Transporter OsAAP4 Contributes to Rice Tillering and Grain Yield by Regulating Neutral Amino Acid Allocation through Two Splicing Variants

چکیده
Abstract Background Amino acids, which are transported by amino acid transporters, the major forms of organic nitrogen utilized higher plants. Among 19 Acid Permease transporters (AAPs) in rice, only a small number these genes have been reported to influence rice growth and development. However, whether other OsAAPs responsible for development is unclear. Results In this study, we demonstrate that OsAAP4 promoter sequences divergent between Indica Japonica , with expression former, produces more tillers grain yield than does . Overexpression two different splicing variants ZH11 significantly increased tillering as result enhancing neutral concentrations Val, Pro, Thr Leu. RNA interference (RNAi) mutant lines displayed opposite trends compared overexpresing (OE) lines. addition, exogenous Val or Pro at 0.5 mM promoted bud outgrowth overexpressing an OsAAP4a variant ZH11, 2.0 enhanced OsAAP4b ZH11. Of note, results protoplast acid-uptake assay showed was specifically accumulated Transcriptome analysis further demonstrated may affect transport metabolism, auxin, cytokinin signaling regulating tillering. Conclusion Our suggested contributes tiller allocation through might potential applications breeding.

Orphan transporter SLC6A18 is renal neutral amino acid transporter B0AT3.
The orphan transporter Slc6a18 (XT2) is highly expressed at the luminal membrane of kidney proximal tubules and displays approximately 50% identity with Slc6a19 (B(0)AT1), which is the main neutral amino acid transporter in both kidney and small intestine. As yet, the amino acid transport function of XT2 has only been experimentally supported by the urinary glycine loss observed in xt2 null mic...
متن کاملAmino acid sensing by enteroendocrine STC-1 cells: role of the Na+-coupled neutral amino acid transporter 2.
The results presented here show that STC-1 cells, a model of intestinal endocrine cells, respond to a broad range of amino acids, including l-proline, l-serine, l-alanine, l-methionine, l-glycine, l-histidine, and alpha-methyl-amino-isobutyric acid (MeAIB) with a rapid increase in the intracellular Ca(2+) concentration ([Ca(2+)](i)). We sought to identify the mechanism by which amino acids indu...
متن کاملThe Rice Peptide Transporter OsNPF7.3 Is Induced by Organic Nitrogen, and Contributes to Nitrogen Allocation and Grain Yield
Nitrogen use efficiency is important for the development of sustainable agriculture. Plants have different transporters to facilitate nitrogen uptake and internal distribution. This study demonstrates that the peptide transporter OsNPF7.3 enhances nitrogen allocation and increases grain yield in rice. OsNPF7.3 is a member of the nitrate transporter 1/peptide transporter family (NPF) and is loca...
متن کاملKnockdown of a nutrient amino acid transporter gene LdNAT1 reduces free neutral amino acid contents and impairs Leptinotarsa decemlineata pupation
A Leptinotarsa decemlineata SLC6 NAT gene (LdNAT1) was cloned. LdNAT1 was highly expressed in the larval alimentary canal especially midgut. LdNAT1 mRNA levels were high right after the molt and low just before the molt. JH and a JH analog pyriproxyfen activated LdNAT1 expression. RNAi of an allatostatin gene LdAS-C increased JH and upregulated LdNAT1 transcription. Conversely, silencing of a J...
متن کاملCysteine Transport through Excitatory Amino Acid Transporter 3 (EAAT3)
Excitatory amino acid transporters (EAATs) limit glutamatergic signaling and maintain extracellular glutamate concentrations below neurotoxic levels. Of the five known EAAT isoforms (EAATs 1-5), only the neuronal isoform, EAAT3 (EAAC1), can efficiently transport the uncharged amino acid L-cysteine. EAAT3-mediated cysteine transport has been proposed to be a primary mechanism used by neurons to ...
